The effects of paclitaxel on nerve conduction velocity and motor unit action potential in spraque-dawley rats

Bu çalışma paklitakselin erişkin erkek Spraque-Dawley ratlarında duyusal ve motor sinir iletim hızları (DSİH ve MSİH) ve motor ünite aksiyon potansiyelleri (MUAP) üzerine olan toksik etkilerini çalışmak için düzenlenmiştir. Ratlar kontrol ve paklitaksel grubu olmak üzere iki eşit gruba ayrılmıştır. Kontrol hayvanlarına 0.5 ml serum fizyolojik, paklitaksel grubuna 6 mg/kg paklitaksel serum fizyolojik içerisinde üç gün boyunca i.p. yolla verilmiştir. Çalışmanın 4, 7, 10 ve 13. günlerinde hayvanlar tartılmış ve pentotal sodyum ile sedate edilerek DSİH, MSİH ve MUAP değerleri belirlenmiştir. Vücut ağırlığı, MUAP ve MUAP süresi değerleri paklitakselden etkilenmemiştir. En fazla etkilenen değişkenler DSİH ve MSİH olmuştur. DSİH ve MSİH değerleri 4.günde paklitaksel grubunda kontrol grubuna göre % 41 ve % 20 azalmıştır (P<0.001 ve P<0.05). Çalışmanın 7, 10 ve 13. günlerinde DSİH ortalaması paklitaksel grubunda kontrole göre sırasıyla % 35, % 20 ve % 33 oranında azalmıştır (P<0.001, P<0.05 ve P<0.01). Buna karşın MSİH ortalama değerleri için gruplar arası farklar onaylanmamıştır. Bağımlı gruplar için t-testi, deney grubunda 10. günde DSİH’nin (P<0.01), 10 ve 13.günlerde MSİH’nin (P<0.05 ve P<0.01) 4. güne göre önemli derecede arttığını göstermektedir. Sonuçlar kemoterapi uygulamaları arasında iki haftalık bir zaman aralığının paklitakselin nöyromuskuler toksik etkilerinin ortadan kalkması için yeterli olmadığını göstermiştir. This study aimed to investigate the toxic effects of paclitaxel on sensory and motor nerve conduction velocities (SNCV and MNCV) and motor unite action potentials (MUAP) in adult male Spraque-Dawley rats. Rats were divided into two equal groups as the control and paclitaxel group. Control animals were injected with 0.5 ml normal saline, while those in paclitaxel group were given 6 mg/kg paclitaxel in normal saline for three days i.p. On days 4, 7, 10 and 13 of the experiment, animals were weighed, and SNCV-, MNCV- and MUAP-values were determined under sedation with Pentothal sodium. Body weights, MUAP and MUAP duration values were not affected by paclitaxel. The most affected variables were SNCV and MNCV, respectively. SNCV- and MNCV-values determined in paclitaxel-group were 41% and 20% less than those of controls on day 4 (P<0.001 and P<0.05, respectively). On days 7, 10 and 13 of the experiment, mean SNCV values of the paclitaxel-group were 35%, 20% and 33% less than those of controls (P<0.001, P<0.05 and P<0.01, respectively). In contrast, MNCV values did not different between groups. The results of t-test for paired groups indicated that in the experimental group mean values of SNCV on day 10 (P<0.01), and MNCV on days 10 and 13 (P<0.05 and P<0.01) increased significantly when compared with corresponding values on day 4. Results indicate that a two-week interval between chemotherapy sections is insufficient for the recovery from the neuromuscular toxic effects of paclitaxel in rats.
