Elektrokimyasal Biyosensörler için Karbon Pasta Elektrot Tasarımı ve Karakterizasyonu
Abstract
Bu çalışmanın amacı grafit tozu ve parafin yağının karıştırılması ile elde
edilen karbon pastanın, 3D yazıcıda bastırılan özel boyutlardaki ABS elektrot
gövdesine yerleştirilmesi ve bu yapının iletken bir tel ile temas ettirilmesi
sonucunda ucuz maliyetli karbon pasta çalışma elektrodunun (KPE)
tasarlanmasıdır. Kitosan-altın nanotanecik-tirozinaz enziminden (K-Gnp-T)
oluşan biyokompozit film ile kaplanan elektrot yüzeyi elektrokimyasal yöntemlerle
karakterize edilmiş ve geliştirilen biyosensörün (KPE*) analitik performansı
fenolik bir bileşik olan kateşolün amperometrik tayini ile test edilmiştir. Ticari
camsı karbon elektrot (CKE) ve bu elektrot yüzeyinin K-Gnp-T biyokompozit filmle
kaplanması ile geliştirilen biyosensör (CKE*) ayrıca karakterize edilmiş ve elde
edilen tüm sonuçlar birbirleri ile karşılaştırılmıştır. Ekonomik açıdan ticari
elektrotlara göre çok daha ucuz bir maliyetle dizayn edilen KPE nin CKE ile benzer
elektrokimyasal özellikler gösterdiği ve KPE*’nın 0.987 korelasyon katsayısı ile
kateşolü yüksek hassasiyette (0.909 A/M) ve CKE*’ ya göre daha geniş doğrusal
aralıkta (0-100 μM) tespit ettiği bulunmuştur. Ayrıca tasarlanan elektrot bir
platform niteliğinde olduğundan farklı birçok sayıda biyosensörün geliştirilmesine
öncülük edebilecek potansiyele sahiptir. The aim of this study is to design the low cost carbon paste electrode
(KPE) as a result of the placing the carbon paste obtained by mixing the graphite
powder and paraffin oil into the ABS electrode body in special dimensions printed
in 3D printer and contacting this structure with a conductive wire. The surface of
the electrode coated with biocomposite film composed of chitosan-gold
nanoparticle-tyrosinase enzyme (K-Gnp-T) was characterized by electrochemical
methods and the analytical performance of the developed biosensor (KPE*) was
tested by amperometric detection of catechol, a phenolic compound. The
commercial glassy carbon electrode (CKE) and the biosensor (CKE*) developed by
coating this electrode surface with a K-Gnp-T biocomposite film were also
characterized and all of the results obtained were compared with each other.
Economically, KPE, which is designed at a much cheaper than commercial electrodes,
shows similar electrochemical characteristics to CKE, and KPE* detected the catechol
at high sensitivity (0.909 A/M) with the correlation coefficient of 0.987 and wider
linear range (0-100 μM) with respect to the CKE*. In addition, the designed electrode
has the potential to lead in the development of many different biosensors due to being a
platform.
